General description
Formyl peptide receptors (FPRs) are G protein coupled receptors critical for our immune response and the cellular activation and or suppression of immune cell activities. N-formyl peptides are produced by the degradation of either bacteria or host cells and serve as chemoattractant agents for white blood cells. There are three known formyl peptide receptors, FPR1, FPR2, and FPR3 all appear to have similar functions. Binding of N-formyl peptides activates neutrophils via the phosphatidylinositol-calcium second messenger system. In some cases binding leads to a down regulation of immune signals such as in the down regulation observed against the proinflammatory signaling agent leukotriene B4. Interestingly, FPRs also have endogenous ligands which appear to function in a developmental fashion and not directly as an immune function. For instance, FPR2’s endogenous ligand is a peptide named Cathelin-related antimicrobial peptide (CRAMP) which is critical for proper dendritic cell maturation. FRP2 is abundantly expressed in the lung, spleen and testis neutrophils. Because of their role in the immune cell activation, FPRs are also being investigated as possible targets for modulating immune caused diseases such as arthritis.
