Specificity
Dopamine D1A Receptor (D1R). The rat D1A receptor is a 446 amino acid, G-protein coupled transmembrane receptor protein (Monsma, 1990; Zhou, 1990; Zhou, 1992; O′Dowd, 1990). The immunogen sequence has 88% sequence similarity with human and 77% with porcine.
Immunogen
A nine amino acid peptide corresponding to the 4th extracellular domain of the rat D1A receptor. The immunogen is absolutely conserved in the mouse D1A receptor and shows 88% homology with human and 77% with porcine and bovine D1A receptors. The peptide shows no significant sequence similarity with other dopamine receptors (D1B, D2-D5).

Application
Detect Dopamine D1A Receptor using this Anti-Dopamine D1A Receptor Antibody, 4th Extracellular Domain validated for use in ELISA, IH & WB.
Immunohistochemistry: 2-10 μg/mL on paraformaldehyde fixed tissue sections. D1R has been localized in various regions of the rat brain (Huang, 1992; O′Connel, 1995)
Western blot: 1-10 μg/mL (Chemiluminescence technique). An antibody made to this peptide has been shown to detect 2 proteins of 50 and 75 kDa in solubilized hippocampal membranes. The 75 kDa band was not visible in striatum (Huang, 1992; O′Connel, 1995). Approximately 100-400 μg of protein/lane should be loaded to visualize the 75 kDa band (Huang, 1992; O′Connel, 1995) that is present in relatively small amounts.
ELISA: 0.5-1.0 μg/mL using 1 μg/mL of control peptide per well.
The immunogen peptide is available (Cat. No. AG266) for use in ELISA or antibody blocking studies. AG266 is not recommended for use as a western blot positive control.
Optimal working dilutions must be determined by end user.
