General description

NEXN (nexilin) is an F-actin binding protein, which is muscle-specific. It resides in adherens junctions.[1] It is predominantly expressed in heart and skeletal muscles. This gene maps to human chromosome 1p31-1p32.

Application

Biochem/physiol Actions

NEXN (nexilin) is responsible for the stabilization of Z-disc and force generation in heart. In smooth muscle cells, its expression is suppressed in response to inflammatory cytokines. It protects vascular smooth muscle cells in injured vessels, by promoting contractility. Variations in this gene are associated with coronary artery diseases in humans.[1] It is a part of focal adhesions, and is thought to play a role in actin-related motility. It is a part of the actin-rich structure of Listeria monocytogenes and enteropathogenic Escherichia coli, and is essential for their invasiveness and infection. Mutations in this gene are also responsible for hypertrophic cardiomyopathy (HCM), which is an inherited disorder characterized by cardiomyocyte hypertrophy, increase in fibrosis of myocardium and increased wall thickness of ventricles. NEXN might also be involved in the formation of stress fibers and focal adhesions, as well as form a part of focal adhesion signaling.
