General description

Uricase is present in many organisms, such as, archaea, bacteria, and eukaryotes. The Gut microbiome is the major source of uricase, a homotetrameric enzyme containing four identical 34 kDa subunits.

Application

Uricase may be used for the determination of uric acid in biological fluids. It may be helpful in the spectrophotometric determination of purines and purine nucleosides.

Biochem/physiol Actions

Uricase catalyzes the oxidation of uric acid to 5-hydroxyisourate, which subsequently forms (S)-allantoin. It is involved in purine metabolism, urate degradation and (S)-allantoin formation The absence of functional uricase in humans has significant physiological and pathophysiological implications. It has been associated with higher serum uric acid levels, leading to an increased risk of conditions related to uric acid crystallization, such as gout. Uricase has emerged as an important therapeutic target for the treatment of conditions related to hyperuricemia, such as gout.
