Phosphatase is a chromogenic substrate for use with alkaline and acid phosphatase ELISA procedures. It is been used for the colorimetric detection of CTB fusion proteins. Produces a soluble end product that is yellow in color and can be read spectrophotometrically at 405 nm. The pNPP reaction may be stopped with 3 N NaOH and read at 405 nm.
Phosphatase substrate has been used as a substrate in alkaline phosphatase assay in serum samples, bone marrow stromal cells and human mesenchymal stem cells (hMSC).
The substrate has been used to determine the alkaline phosphatase (ALP) activity in human osteoblast-like Saos-2 cell lysates. ALP acts as an early marker of osteoblast differentiation that is expressed just before matrix mineralization begins.

Preparation Note
Dissolve tablets to the desired concentration in either of the following buffers: 0.1 M glycine, pH 10.4, with 1 mM MgCl2 and 1 mM ZnCl2 or 1 M diethanolamine, pH 9.8, with 0.5 mM MgCl2. Typically, a pNPP concentration of 1 mg/mL is used.
