General description

Derlin-1 (UniProt Q9BUN8; also known as Degradation in endoplasmic reticulum protein 1, Der1-like protein 1, DERtrin-1) is encoded by the DERL1 (also known as DER1) gene (ORF UNQ243/PRO276; Gene ID 79139) in human. Derlins are integral ER membrane proteins that form homo-oligomers and heteromultimeric complexes with membrane-bound machinery, including the E3 ubiquitin ligase Hrd1 and its binding partner SEL1L, involved in ER-associated degradation (ERAD). Before ERAD substrates can be processed by the cytoplasmic ubiquitin-proteasome system, they need to be translocated across the ER membrane through an AAA ATPase p97/VCP-driven process. Derlins are inactive members of the rhomboid family of intramembrane proteases that mediate the recruitment of AAA ATPase p97/VCP through interaction via their C-terminal SHP boxes. Derlin-1 is proposed to contain four transmembrane segments (a.a. 23-43, 60-80, 106-126, 155-175), three cytoplasmic domains (a.a. 23-43, 60-80, 106-126, 155-175), and two lumenol (a.a. 44-59 & 127-154) domains.

Specificity

Expected to react with both spliced isoforms of human Derlin-1 reported by UniProt (Q9BUN8).

Epitope: C-terminal cytoplasmic domain.
GST-tagged recombinant human derlin-1 C-terminal cytoplasmic fragment.

Application

Immunocytochemistry Analysis: An 1:50 dilution from a representative lot detected Derlin-1 in HUVECs, A431 and HeLa cells.
