Specificity

Clone 1B18 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Transcriptional repressor CTCF. It targets an epitope with in 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from C-terminal region of human Transcriptional repressor CTCF.

Application

Anti-CTCF, clone 1B18 ZooMAb, Cat. No. ZRB1009, is a recombinant Rabbit Monoclonal Antibody that specifically targets Transcriptional repressor CTCF and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CTCF in NIH3T3 and C6 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CTCF in HUVEC, HeLa, A431, and NIH 3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CTCF in human prostate cancer tissue sections.

Target description

Transcriptional repressor CTCF (UniProt: P49711; also known as 11-zinc finger protein, CCCTC-binding factor, CTCFL paralog) is encoded by the CTCF gene (Gene ID: 10664) in human. CTCF is a ubiquitously distributed chromatin binding factor that binds to DNA sequence specific sites. It contains 11 C2H2-type zinc finger regions. CTCF is involved in transcriptional regulation by binding to chromatin insulators and preventing interaction between promoter and nearby enhancers and silencers. CTCF serves as a transcriptional repressor by binding to promoters of vertebrate MYC gene and BAG1 gene. It can also serve as a transcriptional activator of APP. It also plays a critical role in gene silencing over considerable distances in the genome and preferentially interacts with unmethylated DNA, preventing spreading of CpG methylation and maintaining methylation-free zones. When bound to different DNA sequences, it can dimerize to mediate long-range chromatin looping. CTCF undergoes sumolyation on lysine 74 and 689, which contributes to its repressive function on the MYC P2 promoter. Mutations in CTCF gene are known to cause mental retardation characterized by significantly below average general intellectual functioning associated with impairments in adaptive behavior. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
