ELISA Analysis: A representative lot detected LPS from Pseudomonas aeruginosa (It-5) in ELISA applications (Lai, Z., et al. (2005). Vaccine. 23(25):3264-71).

Target description

Pseudomonas aeruginosa is a gram-negative, aerobic, non-spore forming bacteria that causes a variety of infections, particularly in immunocompromised subjects. Following infection, the airway mucosal immune system rapidly develops a pro-inflammatory response, leading to production of inflammatory mediators both in aerial compartment and local tissue initiated by the recognition of pathogen-associated molecular patterns (PAMPs) by specific Toll-like receptors and triggered by the epithelial cells and the resident immune cells. Pathogenesis is mediated by several virulence factors, including motility, adherence to biotic and abiotic surfaces, and secreted toxins. These factors modulate or disrupt host cells signaling pathways, target extracellular matrix, and induce tissue damage. Mouse monoclonal antibody, clone PTA-5385, developed against serotype It-5 directed to LPS O-side chain is opsonic for uptake and killing of bacteria by PMN and is shown to protect against fatal sepsis in mice rendered neutropenic with cyclophosphamide and challenged with homologous P. aeruginosa. Physical form

Purified mouse monoclonal antibody IgG2 in PBS without preservatives.

Reconstitution

1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
