General description

Schiff′s reagent is used for the qualitative or quantitative assay of carbohydrates. Periodic acid-Schiff′s reagent (PAS) stain is broadly applicable in histology for visualizing glycogen, glycoproteins, mucins, and other polysaccharides in tissue and cells.

Application

Schiff′s reagent for aldehydes has been used:
  • in microtiter PAS assay to quantify the amount of secreted mucin in Vibrio cholera infected water
  • to stain the colonies of Clostridium perfringens in PGY agar plates to identify plasmalogen-deficient mutants
  • in periodic acid-Schiff′s reagent assay to quantify carbohydrates in a microtiter plate format
