Specificity

Clone 2L12 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TLR9/CD289. It targets an epitope within 16 amino acids from the N-terminal extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal extracellular domain of human TLR9/CD289.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TLR9/CD289 in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TLR9/CD289 in lysate from EL4 cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected TLR9/CD289 in Daudi cells.

Affinity Binding Assay: A representative lot of this antibody bound TLR9/CD289 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TLR9/CD289 in human liver tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Toll-like receptor 9 (UniProt: Q9NR96; also known as CD289) is encoded by the TLR9 gene (Gene ID: TLR9) in human. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines. TLR9 is a single-pass type I membrane protein that serves as an innate immune sensor that recognizes microbial DNA. It exists as a monomer in the absence of unmethylated cytidine-phosphate-guanosine (CpG) ligand. TLR9 is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form. The mature form contains an extracellular domain (aa 26-818), a short transmembrane domain (aa 819-839), and the cytoplasmic domain (aa 840-1032). The extracellular domain also contains 26 LRR repeat regions (aa 62-772). It is highly expressed in spleen, lymph node, tonsil and peripheral blood leukocytes, especially in plasmacytoid pre-dendritic cell. It is also found in the basolateral region of gastric epithelial cells and high levels are detected in antrum and body mucosa. Five isoforms of TLR9 have been described that are produced by alternative splicing. Ligand recognition of TLR9 is shown to be strictly controlled to prevent unnecessary response to self-derived DNA such as mitochondrial DNA. Ligand-dependent TLR9 trafficking plays a critical role in type I interferon induction in macrophages and plasmacytoid dendritic cells. TLR9 is activated by unmethylated cytidine-phosphate-guanosine (CpG) dinucleotides and acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and the inflammatory response. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
