General description

Exportin-5 (XPO5) belongs to karyopherin-β protein family. The protein shuttles between the nucleus and the cytoplasm.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

Exportin-5 (XPO5) participates in the regulated nuclear export of RNAs and RNA binding proteins. It is involved in nuclear export of interleukin enhancer binding factor (ILF)-3 via RNAs containing minihelix motif in RanGTP dependent manner. Another nuclear export substrate of XPO5 is translation elongation factor 1A (eEF1A). eEF1A binds XPO5 via aminoacylated tRNAs. Interestingly, XPO5 is also involved in nuclear export of pre-miRNAs in RanGTP dependent manner. Depletion of XPO5 results in reduced miRNA levels. XPO5 is important for nuclear export of dicer mRNA, which is required for pre-miRNA maturation. XPO5 is required for nuclear export of 60S subunits in human cells. Single nucleotide polymorphism in XPO5 is associated with increased primary ovarian insufficiency risk. XPO5 is up-regulated in bladder urothelial carcinoma. Silencing XPO5 inhibits cells proliferation and apoptosis in bladder urothelial carcinoma cells.
