Specificity

Clone 1B22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Leucine-rich repeat flightless-interacting protein 2 (LRRFIP2). It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human Leucine-rich repeat flightless-interacting protein 2 (LRRFIP2).

Application

Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LRRFIP2 in A431 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LRRFIP2 in lysate from NIH3T3 cell lysates and recombinant fragment of Human LRRFIP2.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LRRFIP2 in A431 cells.

Affinity Binding Assay: A representative lot of this antibody bound LRRFIP2 peptide with a KD of 5.4 x 10-6 in an affinity binding assay.

Enzyme Immunoassay Analysis (ELISA): A representative lot of this antibody detected recombinant fragment of Human LRRFIP2 in ELISA application.

Target description

Leucine-rich repeat flightless-interacting protein 2 (UniProt: Q9Y608; also known as LRR FLII-interacting protein 2, LRRFIP2) is encoded by the LRRFIP2 gene (Gene ID: 9209) in human. LRRFIP2 is a myeloid differentiation factor 88 (MYD88)-interacting protein with a positive regulatory function in toll-like receptor signaling. It is widely expressed with highest levels observed in heart and skeletal muscle. Five isoforms of LRRFIP2 have been described that are produced by alternative splicing. Isoform 1 is long isoform and other isoforms are considered as short isoforms. It interacts with Dvl3 to activate the canonical Wnt signaling pathway and increases β-catenin/ LEF/TCF-dependent transcriptional activity. Its Dvl3-binding region is localized in amino acids 1-370. Both the amino and carboxyl termini of LRRFIP2 are essential for its function. The amino terminus is responsible for interaction with the central region of Dvl3, which contains the PDZ and DEP domains. Mutant form of LRRFIP2 that contains only the amino terminal acts as a dominant negative form. In resting cells, it shows weak interaction with MYD88. However, following LPS-stimulation, the interaction with MYD is rapidly increased and it returns to basal levels after about six hours of stimulation. Serine 190 and 202 of LRRFIP2 are phosphorylated in response to LPS stimulation. Its interaction with MYD is regulated by LPS-induced phosphorylation of serine 202. LRRFIP2 is also reported to negatively regulate NLRP3 inflammasome activation in macrophages by promoting Flightless-I-mediated caspase-1 inhibition. It binds to NLRP3 via its N-terminal upon NLRP3 inflammasome activation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
