Application

PSI2 is a stable and sustainably prepared P(V) reagent developed by the Baran Lab for the synthesis of phosphorodithioate oligonucleotides. The phosphorodithioate linkages afforded by PSI2 are effectively cleaved by RNAse-H while remaining stable to nucleases, and can be formed rapidly in high yield. The synthetic methodology accessed by PSI2 and other PSI reagents (see ALD00602 and ALD00604) allows at will installation of a greater diversity of linkages to DNA and other nucleotides with comparable performance to existing state-of-the-art P(III) based reagents. The resulting compounds have applications in gene-based therapeutics such as antisense oligonucleotides (ASOs).
