Target description

Lymphocyte antigen 75 (UniProt: O60449; also known as CD205, Ly-75, C-type lectin domain family 13 member B, DEC-205, gp200-MR6) is encoded by the LY75 (also known as CD205, CLEC13B) gene (Gene ID: 4065) in human. It belongs to the to the C-type multilectin family. Its a putative endocytic receptor specific to the uptake and processing of captured antigens for dendritic cells (DC), and is especially known for its expression and presence on the cell surface of CD8+ DCs. CD8+CD205+ DCs are unique in that they seem to be specialized to the uptake of apoptotic cells, in their ability to resist certain viral infections, and their apparent specialization for cross-presentation. CD205/DEC205 is a single-pass type I membrane protein, which is synthesized with a signal peptide (aa 1-27) that is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 28-1666), a transmembrane domain (aa 1667-1691), and a cytoplasmic domain (aa 1692-1722). The large extracellular domain consists of a number of subdomains, including a cysteine rich (CR) domain, a fibronectin type II (FN) domain, and 10 contiguous carbohydrate recognition domains (CRDs). CD205 is predominantly expressed on dendritic cells, colon, spleen, thymus and peripheral blood lymphocytes, and also found in both myeloid and B lymphoid cell lines. Splice variants are known to produce 5 isoforms. Isoforms 2 and 3 are of interest in the study of malignant Hodgkin lymphoma cells or HRS cells. CD205 has been implicated in various diseases, particularly in the context of cancer and autoimmune disorders where aberrant expression or function of CD205 may contribute to dysregulated immune responses. It is also a target of interest in therapeutic drug research for Epstein-Barr virus. (Ref.: Park, C.G., et al. (2012). J Immunol Methods. 377(1-2):15-22).
