Specificity

Application

Quality Control Testing

Evaluated by Flow Cytometry in C57BL/6 mouse splenocytes.

Flow Cytometry Analysis: 1.0 µg of this antibody detected CD226/DNAM-1 in C57BL/6 mouse splenocytes.

Tested Applications

Inhibition: A representative lot inhibited Th1 cell expansion and in the induction of APCs that inhibit T cell activation. (Dardalhon, V., et al. (2005). J Immunol. 175(3);1558-65).

Flow Cytometry Analysis: A representative lot detected CD226/DNAM-1 in Flow Cytometry applications (Dardalhon, V., et al. (2005). J Immunol. 175(3);1558-65; Fang, L., et al. (2009). J Immunol. 182(9):5453-60; Zhang, Z., et al. (2015). J Exp Med. 212(12):2165-82).

Target description

CD226 antigen (UniProt: Q5DW69; also known as Dnam-1, DNAX accessory molecule-1) is encoded by the Cd226 gene (Gene ID: 225825; also known as Dnam-1) in mouse. CD226 is a cell surface glycoprotein that belongs to the immunoglobulin superfamily. It contains 2 Ig-like C2-type domains (aa 19-126 and 297-316) and a transmembrane domain (aa 255-275). It is primarily expressed on the surface of natural killer (NK) cells, T cells, monocytes, and platelets. CD226 plays a crucial role in the regulation of immune responses, particularly in the context of cell-mediated cytotoxicity and immune surveillance. CD226 is involved in the modulation of T cell responses and the regulation of immune tolerance. Dysregulation of CD226 has been implicated in various autoimmune diseases, lupus and cancer, making it an important target for immunotherapy research and treatment development. Physical form

Purified rat monoclonal antibody IgG2b in PBS without azide.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
