General description

Dachshund homolog 1 (UniProt: Q9UI36; also known as Dach1) is encoded by the DACH1 (also known as DACH) gene (Gene ID: 1602) in human. Dach1 is a transcription factor that is involved in regulation of organogenesis. Four isoforms of Dach1 have been reported that are produced by alternative splicing. It is shown to be widely expressed. Isoform 2 is found in brain, heart, kidney, liver, leukocytes and spleen. Isoform 3 is found in liver and heart and isoform 4 is found in spleen. Clone 1F6.1 exhibits reactivity with all four isoforms. Dach1 is reported to act as regulator of SIX1, SIX6, and probably SIX5. It also acts as a a corepressor of SIX6 in regulating proliferation by directly repressing cyclin-dependent kinase inhibitors, including the p27Kip1 promoter. Dach1 is shown to Inhibit TGF-beta signaling through interaction with SMAD4 and NCOR1. It binds to chromatin DNA via its DACHbox-N domain, which forms a structure containing a DNA binding motif similar to that of the forkhead/winged helix domain. PELP1 (proline, glutamic acid, and leucine rich protein 1) serves as a Dach1-binding protein and the carboxyl terminus of Dach1 is required for binding to PELP1. Dach1 is reported to prevent the proliferation of cancerous cells due to being a natural repressor of estrogen receptors (ER) in the normal function of mammary tissue. Specificity

Clone 1F6.1 detects all isoforms of Dach1 in human and murine cells. It targets an epitope within 60 amino acids from the C-terminal region.

Immunogen

GST/His-tagged recombinant fragment coreesponding to 60 amino acids from the C-termnal region of human Dach1.

Application

Anti-Dach1, clone 1F6.1, Cat. No. MABS1805, is a mouse monoclonal antibody that detects Dach1 in human and murine cells and has been tested for use in Immunohistochemistry (Paraffin) and Western Blotting.
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected Dach1 in human bone marrow and human colon cancer tissues.

Western Blotting Analysis: 0.5 µg/mL from a representative lot detected Dach1 in Neuro-2a cell lysate.
