Specificity

Clone 3N3 is a Rabbit recombinant monoclonal antibody that specifically detects Calcium uniporter protein, mitochondrial (MCU). It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Calcium uniporter protein, mitochondrial (MCU).

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MCU in A549 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MCU in PANC-1 and NIH3T3 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound MCU with a KD of 2.0 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MCU in A549 cells.

Target description

Calcium uniporter protein, mitochondrial (UniProt: Q8NE86; also known as HsMCU, Coiled-coil domain-containing protein 109A, MCU) is encoded by the MCU (also known as C10orf42, CCDC109A) gene (Gene ID: 90550) in human. MCU is a multi-pass inner mitochondrial membrane calcium uniporter that mediates calcium uptake into mitochondria. It is synthesized with a transit peptide (aa 1-50), which is subsequently cleaved off to generate the mature MCU protein that has two mitochondrial matrix domains (aa 51-233 and 284-351), two transmembrane domains (aa 234-256 and 266-283) and one mitochondrial Intermembrane domain (aa 257-265). Its activity is regulated by the mitochondrial calcium uptake 1 (MICU1) and MICU2. MICU1 is a single pass membrane protein that helps to stabilize the entire mitochondrial calcium uniporter complex. It also limits the amount of calcium that enters the cell during low concentrations of calcium. Also, it functions alongside MCU to keep the accumulated calcium inside the mitochondrial matrix. MICU2 works with MICU1 and MCU to reduce the amount of calcium coming into the matrix. At low calcium levels MCU activity is down-regulated by MICU1 and MICU2 and at higher calcium levels MICU1 increases MCU activity. Synaptic activity is also reported to transcriptionally represses MCU gene, via a mechanism involving the nuclear Ca2+ and CaM kinase-mediated induction of Npas4, resulting in the inhibition of NMDA receptor-induced mitochondrial Ca2+ uptake and preventing excitotoxic death. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
