Specificity

Clone 1B12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Sodium/bile acid cotransporter (SLC10A1). It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human Sodium/bile acid cotransporter (SLC10A1).

Application

Quality Control Testing

Evaluated by Western Blotting in Human liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SLC10A1 in Human liver tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected recombinant Human SLC10A.

Enzyme Immunoassay Analysis (ELISA): Serial dilutions from a representative lot detected recombinant Human SLC10A1 protein.

Target description

Sodium/bile acid cotransporter (UniProt: Q14973; also known as Cell growth-inhibiting gene 29 protein, Na(+)/bile acid cotransporter, Na(+)/taurocholate transport protein, Sodium/taurocholate cotransporting polypeptide, Solute carrier family 10 member 1) is encoded by the SLC10A1 (also known as NTCP, GIG29) gene (Gene ID: 6554) in human. SLC10A1 is a multi-pass membrane glycoprotein with eight transmembrane domains. It is expressed exclusively on the basolateral membrane of hepatocytes and serves as a major high-affinity Na+-dependent transporter of conjugated bile salts from plasma into hepatocytes. It plays a key role in the enterohepatic circulation of bile salts necessary for the solubilization and absorption of dietary fat and fat-soluble vitamins. About 75% of the conjugated bile acids and about 50% of unconjugated bile acids are imported via this transporter. The inward Na+ gradient necessary for this process is maintained by a Na+/K+ ATP pump. This process is shown to be extremely efficient, with 75% to 90% first-pass clearance of bile acids from the sinusoidal blood. SLC10A1 exhibits broad substrate specificity and transports various non-bile acid organic compounds as well. Mutations in SLC10A1 gene are reported to cause familial hypercholanemia 2 (FHCA2) that is characterized by persistently higher plasma levels of conjugated bile salts that is apparent from infancy associated with fat malabsorption and impaired absorption of fat-soluble vitamins. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
