Specificity

Clone 1F2 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PIK3R3. It targets an epitope within 24 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal half of human PIK3R3.

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PIK3R3 in L6 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PIK3R3 in lysates from Neuro-2a and MDA-MB-435 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PIK3R3 in human cerebral cortex tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with PIK3R3 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PIK3R3 in SH-SY5Y cells.

Target description

Phosphatidylinositol 3-kinase regulatory subunit gamma (UniProt: Q92569; also known as PI3-kinase regulatory subunit gamma, PtdIns-3-kinase regulatory subunit gamma, Phosphatidylinositol 3-kinase 55 kDa regulatory subunit gamma, PI3-kinase subunit p55-gamma, PtdIns-3-kinase regulatory subunit p55-gamma) is encoded by the PIK3R3 gene (Gene ID: 8503) in human. PI3K phosphorylates phosphatidylinositol and similar compounds, which then serve as second messengers in growth signaling pathways. PI3K is composed of a catalytic and a regulatory subunit. The protein encoded by this gene represents a regulatory subunit of PI3K. The encoded protein contains two SH2 domains (SH2 1 and SH2 2) through which it binds activated protein tyrosine kinases to regulate their kinase activity. During insulin stimulation, it also binds to IRS-1. PI3K is expressed in various tissues, with notable high levels in brain and testis. It is also found in lower levels in adipose tissue, kidney, heart, lung and skeletal muscle. It is predominantly localized in the cytoplasm and can translocate to the plasma membrane upon activation. Three isoforms of PI3K have been described that are produced by alternative splicing. Dysregulation of the PI3K signaling pathway, often due to mutations in PIK3R3 or its associated components, can lead to uncontrolled cell growth and survival, contributing to tumorigenesis. It is indicated that PIK3R3 promotes colorectal cancer metastasis by positively regulating cell migration and invasion. PIK3R3 also plays a pivotal role in ovarian cancer development and its expression is upregulated in ovarian cancer as well as ovarian cancer stem cells. Inhibition of PIK3R3 reduced the spheroid size of ovarian cancer stem cells. PIK3R3 is suggested to be a useful prognostic marker for ovarian cancer and can therefore be exploited for developing therapeutic strategies for this cancer type. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
