General description

Specificity

Clone 1H11 is a ZooMAb® rabbit recombinant monoclonal antibody that detects p57Kip2. It targets an epitope within 25 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human p57Kip2.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected p57Kip2 protein in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected p57Kip2 in lysates from dexamethasone (100 nM), treated A549 cells, mouse kidney tissue, rat adrenal tissue, and human kidney tissue; and a 1:10,000 dilution from Jurkat cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected p57Kip2 in human placenta tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected p57Kip2 in Jurkat cells and MCF7 cells.

Affinity Binding Assay: A representative lot of this antibody bound with p57Kip2 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Flow Cytometry Analysis: 1 µg from a representative lot detected p57Kip2 in one million MCF7 cells.

Target description

Cyclin-dependent kinase inhibitor 1C (UniProt: P49918; also known as Cyclin-dependent kinase inhibitor p57, p57Kip2) is encoded by the CDKN1C (also known as KIP2) gene (Gene ID: 1028) in human. p57Kip2, is a member of the Cip/Kip family of cyclin-dependent kinase (CDK) inhibitors. It is characterized by an N-terminal domain that is crucial for its inhibitory activity towards CDKs, particularly CDK2 and CDK4. p57Kip2/CDKN1C functions as a potent inhibitor of cell cycle progression. By binding to and inhibiting CDKs, it plays a critical role in regulating the G1 phase of the cell cycle. p57Kip2 also participates in other cellular processes, including apoptosis and senescence. CDKN1C is expressed in a variety of tissues. Its expression is tightly regulated by genetic and epigenetic mechanisms, including imprinting, where typically only the maternal allele is expressed. The expression of CDKN1C can be influenced by developmental cues and stress signals. Mutations or dysregulation of CDKN1C are associated with developmental disorders such as Beckwith-Wiedemann syndrome, a condition characterized by overgrowth and an increased risk of childhood tumors. Additionally, alterations in CDKN1C expression or function can contribute to cancer development, as its role in inhibiting cell cycle progression is critical for preventing uncontrolled cell division. Loss of CDKN1C function can lead to unchecked cell proliferation, a hallmark of cancer. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
