Specificity

Clone 1B5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects High affinity immunoglobulin gamma Fc receptor I (CD64). It targets an epitope within 21 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the extracellular domain of human CD64.

Application

Quality Control Testing

Evaluated by Western Blotting in Human tonsil tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD64 in Human tonsil tissue lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound CD64 peptide with a KD of 3.6 x 10-8 in an affinity binding assay.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected CD64 in one million U937 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD64 in U937 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

High affinity immunoglobulin gamma Fc receptor I (UniProt: P12314; also known as IgG Fc receptor I, Fc-gamma RI, FcRI, Fc-gamma RIA, FcgammaRIa, CD64) is encoded by the FCGR1A (also known as FCG1, FCGR1, IGFR1) gene (Gene ID: 2209) in human. CD64 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-15), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 16-292), a transmembrane domain (aa 293-313), and a cytoplasmic domain (aa 314-374). Within its extracellular domain are three Ig-like C2 type domains. Its expression has been reported in monocytes/macrophages. Expression of CD64 distinguishes conventional dendritic cells from monocyte-derived dendritic cells. It serves as a high-affinity receptor for the Fc domains of IgG (Fc R) and plays a key role in linking the humoral and the cellular arms for the immune system. It binds human IgG1, IgG3, and IgG4 with a high affinity, but does not show any affinity for IgG2. It functions in both innate and adaptive immune responses. Along with the low affinity receptor (Fc RII), it mediates a variety of biological responses, including endocytosis, phagocytosis, antibody-directed cellular cytotoxicity, and cytokine release. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
