Specificity

Clone 2B18 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TBX4. It targets an epitope within 19 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal half of human TBX4.

Application

Quality Control Testing

Evaluated by Western Blotting in Caco-2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TBX4 in Caco-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TBX4 in nuclear extracts from HepG2 and U-2 OS cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected TBX4 in one million Caco-2 cells.

Affinity Binding Assay: A representative lot of this antibody bound with TBX4 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TBX4 in THP-1 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TBX4 in human liver tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

T-box transcription factor TBX4 (UniProt: P57082; also known as T-box protein 4) is encoded by the TBX4 gene (Gene ID: 9496) in human. The TBX4 gene is a member of a phylogenetically conserved family of genes that share a common DNA-binding domain, the T-box. T-box genes encode transcription factors involved in the regulation of developmental processes. This gene is the human homolog of mouse Tbx4, which is closely linked to Tbx2 on mouse chromosome 11. Similarly this gene, like TBX2, maps to human chromosome 17. Expression studies in mouse and chicken show that Tbx4 is expressed in developing hindlimb, but not in forelimb buds, suggesting a role for this gene in regulating limb development and specification of limb identity. This transcriptional regulator also plays an essential role in the organogenesis of lungs, pelvis and the development of the cardiovascular system. TBX4 is primarily localized in the nucleus and is predominantly expressed during embryonic development, particularly in the developing limbs and various mesodermal tissues. In adults, its expression is more restricted, being found in tissues such as the lung, placenta and prostate. Mutations in TBX4 gene can lead to conditions such as small patella syndrome, characterized by absent or hypoplastic patellae and limb malformations. TBX4 variants have increasingly been associated with a variety of clinical and histopathological phenotypes, including lethal developmental parenchymal lung diseases such as not only acinar dysplasia in neonates, but also less outspoken parenchymal lung diseases in children and adults. Heterozygous variants in TBX4 are the second leading genetic cause of pulmonary arterial hypertension (PAH). This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
