General description

Ni-NTA functionalized gold nanoparticles are suitable for binding of proteins with a C-, or N-terminal 6X His tag. These gold nanoparticles allow for rapid generation of gold conjugates for subsequent use in downstream assays.

Ni-NTA functionalized gold nanoparticles are more than 95% spherical and have uniform size distribution (CV <12%).

Application

Ideal for development of gold conjugates for use in applications such as blotting, lateral flow assays, microscopy, and transmission electron microscopy (TEM).

Storage and Stability

When stored for a long period of time gold nanoparticles may sediment at the bottom of the vial, which is especially prominent for larger particle sizes. Prior to use, re-suspend the sedimented particles by thorough mixing until a homogenous solution is obtained.

A structure-based approach was pursued in designing novel bisphosphonate inhibitors of the human farnesyl pyrophosphate synthase (hFPPS). Preliminary SAR and structural evidence for the simultaneous binding of these inhibitors into the isopentenyl pyrophosphate (IPP) and the geranyl pyrophosphate (GPP) substrate

Isoprenoids, most of them synthesized by prenyltransferases (PTSs), are a class of important biologically active compounds with diverse functions. The mint geranyl pyrophosphate synthase (GPPS) is a heterotetramer composed of two LSU·SSU (large/small subunit) dimers. In addition to C(10)-GPP, the

Farnesylation of the gamma-subunit of the retinal G-protein, transducin (Talpha/Tbetagamma), is indispensable for light-initiated signaling in photoreceptor cells. However, the farnesyl-mediated molecular interactions important for signaling are not well understood. To explore this issue, we created a functional Tbetagamma analogue

A mix-and-read FlashPlate (PerkinElmer, Waltham, MA) assay for the enzyme farnesyl pyrophosphate (FPP) synthase (FPPS) was developed to rapidly measure both steps in the synthesis of FPP from dimethylallyl pyrophosphate (DMAPP). The assay used either DMAPP or geranyl pyrophosphate (GPP)

Terpenes (isoprenoids), derived from isoprenyl pyrophosphates, are versatile natural compounds that act as metabolism mediators, plant volatiles, and ecological communicators. Divergent evolution of homomeric prenyltransferases (PTSs) has allowed PTSs to optimize their active-site pockets to achieve catalytic fidelity and diversity.
