General description

TRANCE (TNF-related activation-induced cytokine), also called RANKL (RANK ligand), OPGL (Osteoprotegrin ligand), and ODF (Osteoclast differentiation factor) is a member of tumor necrosis factor superfamily that is expressed on the surface of osteoblasts and is released by activated T cells. When TRANCE binds to its receptor RANK it stimulates the formation, activation, survival and adherence of osteoclasts that increases the bone resorption. These effects are inhibited when RANKL binds to OPG that leads to decrease in bone resorption process and increase in bone volume and density. The TRANCE (RANKL)/RANK/OPG collectively activates NF-κB, JNK, Akt, p38, ERK, NFATc1 pathways. This axis is vital for maintaining the bone density, dendritic cell survival, B cell differentiation, mammary gland development. This signaling pathway maintains the osteoclast biology and is a therapeutic target for osteoporosis. Monoclonal Anti-TRANCE antibody recognizes human TRANCE (~27 kDa). This antibody demonstrates 5% cross reactivity with recombinant mouse TRANCE.

Application

Anti-TRANCE antibody may be used for immunohistochemistry of paraffin-embedded tissue sections at a working concentration of 8-25 μg/ml. For ELISA a concentration of 0.5-1.0 μg/ml is recommended. The antibody is also suitable for neutralization reactions (ND50 is 0.02-0.06 μg/ml).

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline with 5% trehalose.
